The support for Windows Azure was not present in 4.x versions, it was added in 5.5 R2, so you will definitely need to upgrade.

I suppose you're currently running the Web Site project, correct? Because even if you upgrade, you would still need to convert your application to Azure project and that would be lot more difficult than migrating your data to freshly installed Azure project...

One possible way would be to upgrade the website all the way to version 7, export the website, then install fresh Azure project (v7) and import the site into this instance.

If you are using any code customizations, you will need to redo them in the fresh project as well. We would need to know further details to give you more accurate advice on how to proceed.
